A human antibody recognizing a conserved epitope of H5 hemagglutinin broadly neutralizes highly pathogenic avian influenza H5N1 viruses.

This set of residues is found conserved among all the (sub)clades of the H5N1 subtype except for subclade 7.2 (such as A/Chicken/Vietnam/NCVD-016/08) where Serine 121 is subsituted with Tyrosine.
The ability of the epitope specific mAb to neutralize pseudotyped lentiviral infection of cells was assessed. Mutation of each epitope residue (P118, S121, K161, Y164, T167) of this strain generated variants more resistant to neutralization than the WT pseudovirus.
