Neutralization of macrophage migration inhibitory factor (MIF) by fully human antibodies correlates with their specificity for the beta-sheet structure of MIF.
|
| description: |
The epitope sequence was deduced.
Administration of the epitope-specific antibody suppressed the effector phase of delayed-type hypersensitivity, as indicated by reduced ear swelling after re-exposure to DNFB. The antibody also inhibited sensitization of the immune system, as shown by reduced adoptive transfer of contact hypersensitivity to DNFB in naive mice receiving donor cells from antibody-pretreated, DNFB-sensitized mice. BaxM159 is a higher affinity variant of BaxG03 that was generated by through complementarity determining region 1 and 2 and light chain shuffling and selection by phage display with limiting concentrations of antigen.
