The phage displayed epitope inhibited the binding of Bla g 2 to anti-Bla g 2 mAb 7C11. MAb 7C11 was used to select the epitope from a random 12-mer phage display library by three rounds of panning. Phage displaying the epitope sequence or the same sequence with an Glu to Asp substitution at position 8 were the only clones selected. The authors mapped the epitope sequence to patches on the surface of the Bla g 2 structure centered on residues Pro61 and Arg83, which overlap with the epitope determined from the crystal structure.
