The epitope is the terminal trisaccharide from blood group A antigen, and is shared among the three type A antigens.
Human Abs (IgG + IgM + IgA) from blood type B donors reacted with the epitope strongly, while a low but detectable reactivity was found in A blood type sera. Affinity purification of the epitope-specific Abs showed that the B-type alloantibodies do not cross-react with group A terminal minimal disaccharide and requires at least the epitope trisaccharide, while the A-type affinity purified epitope-specific auto-antibodies are reactive with the minimal disaccharide. This type of reactivity is shown to be of true auto-antibodies, not cross-reactive to B-type antigens, as it doesn't disappear when the serum was adsorbed to B-type trisaccharide antigen.
