The discontinuous epitope residues are located in the DI and DI-DII hinge regions of the Dengue virus 1 WP 74 envelope protein.
The epitope-specific mAb was shown to neutralize homologous DENV, but not DENV2-4. The epitopic residues were mapped by sequence analysis of viral escape mutants. Binding was not observed to recombinant E protein, EDIII, nor premembrane protein, suggesting the epitope is preserved only on full viral particles.
