The heavy chain constant region of human IgG3 was linked to the variable heavy chain region of anti-dansyl murine mAb 27-44. The antibody antigen was expressed with a chimeric light chain made of human kappa constant region linked to the variable light chain region of the same murine mAb. The use of mutants and exon-shuffled chimeric IgG4 and IgG3 molecules allowed for the determination of the epitopic residue as involved in the recognition by a monoclonal Rheumatoid Factor (RF) antibody obtained from Rheumatoid Arthritis patients.
