The epitope-specific mAb recognized the antigen in brain sections of human Alzheimer tissue. Strong immunolabeling of A beta plaques was seen. Three-dimensional reconstruction of confocal microscopy images unequivocally revealed the presence of intracellular mAb-positive puncta, thus sustaining their internalization. EM analysis of immunogold-labeled mC99(1–7) in pretreated HEK cells confirmed the localization of these mAbs in multivesicular bodies.
