| name: |
Hemostatic effect of a monoclonal antibody mAb 2021 blocking the interaction between FXa and TFPI in a rabbit hemophilia model.
|
| description: |
The epitope residues are identified as E8, E9, D10, P11, R15, Y17, T19, Y21, F22, N24, Q26, Q29, C30, E31, R32, K34, N41, E46, and L48, according to BPTI numbering, in the text and Figure 1 of the reference.
Administration of epitope-specific mAb 2021 before induction of hemophilia in rabbits reduced blood loss in a dose-dependent manner. Administration of mAb 2021 4 or 7 days before induction of transient hemophilia reduced blood loss, but mAb 2021 lost its effectiveness when administration 10 days in advance. MAb 2021 also reduced ongoing bleeding in rabbits treated 5 min after induction of cuticle bleeding or pretreated by s.c. injection 24 h in advance.
