Protein extracts of Arabidopsis wild type and the N-glycosylation mutants were tested by Western-blot with the epitope-specific antibodies. The labeling of glycoproteins from mutants lacking Xylosyl transferase activity was markedly reduced compared to wild-type. Enzymatic treatment of A. thaliana protein extract with glycosidases removing residues different from Xylose also affected the recognition of the glycoprotein, implying the conformation and accessibility to recognition of the Xylose residue is affected by the presence other N-glycan residues.
