The epitope was deduced.
The epitope residues recognized by the platelet-associated (PA) anti-alphaIIbß3 Abs from ITP patients were determined by analysis of binding to human-mouse chimera alphaIIbß3 molecules with small sections of the N-terminal half of the human molecule exchanged with the corresponding mouse sequence. Exchange of the mouse sequence for the human epitope (W4:4-1) reduced binding of PA Abs from 4 of 15 ITP patients. In addition, mouse alphaIIbß3, with the human W4:4-1 sequence exchanged in, was recognized similar to WT human alphaIIbß3 by 3 of the 4 PA anti-alphaIIbß3 Abs.
