Epitopic residues recognized by serum IgE from allergic patients were identified by analysis of binding to Blo t 21 with single alanine substitution at each of 17 charged residues. Mutations that caused a >20% reduction in IgE binding in more than 50% of the patients' sera were: E89A (11 of 12 patients), D79A (8 of 12), D96A (7 of 12), K107A (7 of 12), and E84A (6 of 12). Mutation E74A reduced IgE binding in 4 of 12 patient sera and was also identified as an IgE epitope in Blo t 5. IgE binding was further reduced to Blo t 21 containing mutations at two to four of the residues E74, D79, E89, and D96.
