The Aß1-42 peptide inhibited the binding of epitope-immunized serum from binding to the Aß1-42 peptide in a competitive ELISA. Direct ELISAs showed that immunization with a vaccine consisting of a dense array of the epitope on liposomes induced epitope-specific IgG responses totally independent of T-cell help. Independency was confirmed in mice lacking T cells and in mice deficient for MHC class II, CD40L, and CD28. TLR4 and TRIF are required for the induction of epitope-specific IgG response.
