Mutational analysis of the PTP IA-2 region revealed a significant reduction of binding of T1D patient sera when residue W799 and C909 were substituted with alanine or serine, respectively. This suggests that these residues are critical for binding. Pretreatment with N-ethylmaleimide caused little additional reduction of binding by these sera. Interestingly, serine substitutions at C623, C717, C726 and C820 caused modest increases in binding of patient sera.
