Following the second round of panning, the presence of an enriched population of phage-displayed scFvs demonstrating binding to Gal-alpha-(1,3)-Gal-BSA was confirmed by direct ELISA. From 74 random picked positive clones, 6 were sequenced and 4 unique sequences were isolated. Clone A4 was expressed as soluble scFvs and confirmed by direct and competitive ELISA to bind specifically to the epitope, and minimally to other carbohydrates bound to BSA or HSA, except for those containing the epitope. The scFv was used to detect and quantify the epitope content in several glycoproteins.
