Several shortenings and alanine- and conservative-variant peptides from the originally determined epitopic region ECTGLAWEWWR from PLY protein, conserved in many bacterial cholesterol-dependent cytolysins (CDCs), were tested as inhibitors of mAb PLY-5 neutralising activity in PLY-mediated haemolysis. The minimal epitope was deduced to be WEWWR, with residues W3 and R5 essential for recognition.
