The epitope was deduced.
The epitope-specific mAbs reacted by flow cytometry with CHO cells stably expressing the TSHR ECD tethered to the plasma membrane by a GPI anchor. For mAb 3BD10 the estimated Kd based on the fluorescence with increasing concentrations of mAb was calculated at 1.2 nM. The region CSSPPC, corresponding to loop 1, was found to affect binding to the protein by mutation to residues CPEPCD as found on the FSH receptor, but not when individual residues were deleted, suggesting the loop 1 conformation affects recognition of the loop 2-specific mAbs.
