The epitope is located on integrin beta-1. The epitope residues were calculated from [PDB: 3VI3] as the antigen residues at 4 Å
atomic distance from the antibody. The epitope calculated from [PDB: 3VI4] contained one additional residue, K87.
The epitope of SG/19 Fab on the headpiece fragment of alpha5ß1 integrin was determined from the crystal structure of the complex. The hexapeptide GRGDNP was soaked into the crystal. The electron density of the ligand peptide relative to integrin suggested that the occupancy of the peptide was not 100%. The complex was crystallized in the presence of physiological concentrations of Mg2+ and Ca2+. Two alpha5beta1–Fab complexes contained in one crystallographic asymmetric unit were essentially identical. Therefore, only one complex discussed in the paper (chains A, B, E, and F) was curated.
