Structural insights into the down-regulation of overexpressed p185(her2/neu) protein of transformed cells by the antibody chA21.
description:
The epitope-specific chA21 mAb bound to p185(her2/neu) on the surface of the human mammary cancer cell line SK-BR-3 and down-regulated expression of p185(her2/neu). The scFv chA21 did not down-regulate p185(her2/neu), indicating the requirement for bivalency. MAb chA21 also down-regulated expression of EGFR and ErbB3, which form heterodimers with p185(her2/neu). MAb chA21 was also shown to form high molecular weight complexes with a p185(her2/neu) ectodomain-human Fc fusion protein.
