The affinity of epitope-specific mAb 4E11 for binding the extracellular domain III of DENV-2 was determined by surface plasmon resonance. It was also determined by competition ELISA (KD = 5.2 nM). The mAb 4E5A, engineered for improved affinity, showed a 21-fold increase in affinity for EDIII of DV2 by SPR and 9-fold increase by competition ELISA.
