The affinity of the epitope-specific antibody GL-VRC01 for the HIV gp120 outer domain construct eOD-GT6 was determined by surface plasmon resonance. The affinity of the mature antibody VRC01 for eOD-GT6 was KD = 2 nM. The eOD-GT6 construct was based on an engineered outer domain lacking the inner domain, V1, V2, V3 regions and ß20/21 with 8 additional mutations introduced, including removal of glycosylation sites. Six of these mutations improved the affinity. The construct contained 10 mutations relative to HIV HxB2 gp120.
