The toxicity of antiprion antibodies is mediated by the flexible tail of the prion protein.

The epitope-specific antibody POM8 and POM9 induced loss of cerebellar granule cells in PrPC -overexpressing cerebellar organotypic cultured slices (COCS). Neurotoxicity required the expression of Prp, as Prnp(o/o) COCS were not affected. Monovalent (Fab or scFv) or divalent antibodies (IgG or Fab 2) was neurotoxic indicating that neurotoxicity did not require crosslinking or antibody effector function or complement. Neurotoxicity did require the flexible tail domain of PrP, as COCS expressing a PrP deletion mutant lacking residues 32-93 were resistant to POM neurotoxicity.
