The affinity of Fv 447-52D for the linear V3 MN epitope was determined by isothermal calorimetry. The ka = 0.049 1/M. Mutation of antibody heavy chain residues, Y100T, W133A, or D95A or light chain residues, W91A or W96A, reduced the affinity. Mutation of position R9Q in the epitope reduced the affinity 230-fold, whereas mutation H4R resulted in a 2.3-decrease in binding affinity.
