The affinity of epitope-specific H5M9 IgG for binding to H5 HA of Influenza A virus (A/Hong Kong/156/97) was determined by bio-layer interferometry using an Octet Red instrument. For the Fab', KD = 0.9 nM. Binding of H5M9 Fab' protected the HA from degradation by trypsin at pH 4.9, suggesting that antibody binding inhibits the pH-induced conformational changes in HA needed for virus function.
