The epitope-specific MAb492.1 inhibited the sulfhtdryl oxidase activity ofQSOX1 secreted from confluent fibroblasts. The IC50 = 43 nM in an assay for inhibition of RNase oxidation. The apparent inhibitory constant was determined to be Ki = 0.9 nM in an oxygen consumption assay. In a physiological assay, MAb492.1 inhibited epithelial adherence to fibroblasts and inhibited tumor cell migration on fibroblast monolayers. The Fab492.1 and scFv492.1 fragments also inhibited RNase oxidation with IC50 = 100 nM and IC50 = 250 nM, respectively.
