The epitope residues are calculated from the PDB structures [PDB: 4LDE, 4LDL and 4LDO] as the antigen residues at 4 Å
distance from the antibody. The epitope for PDB 4LDO does not include contact residues T1068 (T68) and A1134 (A134) and has an additional contact residue A1265, which is mismatched to C265 in the Genbank sequence.
The epitope of nanobody Nb6B9 on the ß2AR-T4 lysozyme fusion protein was determined from the crystal structure, solved by molecular replacement. In the fusion protein, human ß2AR was fused to an N-terminal T4 lysozyme. Crystals of ß2AR-T4L bound to high-affinity catecholamine agonist hydroxybenzylisoproterenol(HBI) in complex with Nb6B9 were grown in lipidic cubic phase.
