The epitope sequence was deduced by virus escape mutants.
The epitope-specific mAb was able to neutralize infectivity of DENV-1. Plaque reduction neutralization of infective titers were determined on Vero cells. The epitope-specific mAb did not display enhancing activity even at very low concentrations, measured by Dengue antibody-dependent enhancement (ADE) assay. Dose-dependent antibody activity patterns generated using K562, U937, and HL60 cells were comparable. The mAb engineered with IgG3 showed neutralizing activities without any enhancing activities, enhancing activities appeared upon substitution to IgG1 in both the presence and the absence of complement and to IgG2a and IgG2b in the absence of complement.
