The epitope-specific mAb 132A reacted with the fragment 1-161. Truncation analysis revealed reactivity with smaller fragments (1-161, 1-140, 1-130, 1-120, 1-110 and 1-90), but not with 1-77. Further mutational analysis of the alpha subunit N-terminus showed that the epitope consisted of two segments, aa1-12 and aa65-79. A third segment (aa110-115) was shown to play a role in the stabilization of the folding of the center MIR segment.
