The epitope was deduced.
The epitope of mAb Ab64 on rGroEL from strain LVS was mapped by hydrogen/deuterium exchange-mass spectroscopy (DXMS). The region of residues 329-350 showed reduced deuterium exchange in the presence of the antibody. This region included residue K344, but not residue Y476, which were found to be epitope residues by point mutation analysis. The authors suggest that mutation Y476E, which abolished Ab64 binding exerts an allosteric effect on the structure of the epitope.
