The epitoipe inhibited the binding of the mAb F22-4 to Shigella flexneri 2a LPS. The epitope did not significantly inhibit the binding of SF2a-specific protective mAbs A2-1 (IgG2a), D15-7 (IgG1), C1-7 (IgG3) or E4-7 (IgG2b). The authors conclude that while the 6D′-acetate has no major influence on Ab recognition, the 3A-O-acetyl group displays a striking mIgG-specific effect on Ab binding. Only mAb F22-4 is able to recognize diversely O-acetylated segments of the SF2a O-Ag chain. None of the two acetates present on the SF2a O-Ag adds significantly to broad Ab recognition.
