The epitoipe inhibited the binding of the mAb F22-4 (IgG1) to Shigella flexneri 2a LPS. The epitope also inhibited the binding of SF2a-specific protective mAbs A2-1 (IgG2a; IC50 = 2 µM) , D15-7 (IgG1; IC50 = 21 µM) , C1-7 (IgG3; IC50 = 22 µM) and E4-7 (IgG2b; IC50 = 7 µM). The authors conclude that while the 6D′-acetate has no major influence on Ab recognition, the 3A-O-acetyl group displays a striking mIgG-specific effect on Ab binding. Only mAb F22-4 is able to recognize diversely O-acetylated segments of the SF2a O-Ag chain. None of the two acetates present on the SF2a O-Ag adds significantly to broad Ab recognition.
