Immunization with NP-alphaGalCer stimulated higher titers of anti-NIP IgG and IgM than immunization with NP-KLH in wild type C57BL/6 mice. Immunization of CD1d-/- and Jalpha18-/- mice with NP-alphaGalCer did not stimulate an anti-NIP response, indicating the requirement for iNKT cells. CD4 was not required for a response. Immunization with NP-alphaGalCer preferentially induces IgM, IgG2c and IgG3 anti-NIP. Immunization with NP-KLH stimulated preferentially IgG1 and IgG2b anti-NIP titers.
