The residue numbering in the manuscript refers to the positions in the HA1 (H18, H38, Q40, S291, M292) and HA2 (V18, D19, G20, W21, K38, T41, Q42, I45, T49, V52, N53, I56) chains cleaved from the hemagglutinin precursor, and follows the structure of H3 in [PDB 2HMG].
The epitope specific mAb was expressed as a scFV-Fc construct or was reconstituted as a full human IgG1 from the scFV construct. It showed neutralizing activity towards the H5 subtype, clade 0 (A/HongKong/483/97), clade 1 (A/Viet Nam/1203/2004 and A/Thailand/2-SP-33/2004) and clade 2.1 (A/Indonesia/5/2005) viruses, as well as other group 1 viruses (H1N1, H2N2, H6N1, H6N2, H8N4 and H9N2 viruses, and H11-pseudotyped VLPs). It failed to neutralized group 2 (H7 and H3 subtype viruses). Analysis of the neutralizing activity with H5-pseudotyped VLPs or in cells transfected with the HA molecule showed that the nAb inhibits membrane fusion and syncitia formation rather than receptor binding.
