Epitope (DQ2.5-glia-alpha2)-specific T cells responded to the epitope presented by A20 cells transfected with DQ2.5 or by A20-DQ2.5-IgD cells, as measured by murine IL-2 release. A20-DQ2.5-IgD are cells expressing both HLA-DQ2.5 and the TG2-binding IgD 679-14-E06. The cells also responded to A20-DQ2.5-IgD cells loaded with preformed complexes of TG2 with non-deamidated 33mer, which becomes deamidated during complex formation. This indicates a role for the TG2-specific BCR in internalization and processing of the complexes for presentation and recognition by T cells.
