The affinity of epitope-specific HCV1406 TCR for the epitope/A2 complex was determined by surface plasmon resonance. The affinity to the p1 Lys→Ala (K1A) NS3 variant was reduced to KD ≥170 μM. Mutation of polymorphic residues Arg65 and Lys66 in the HLA-A2 alpha chain drastically reduced binding by the HCV1406 TCR. Mutation of other polymorphic sites, Ala69, Val76, and Gly79, also reduced binding to various extents..
