The binding interactions of human TCR molecule A6 to the complex of HLA-A2/epitope were examined by Surface plasmon resonance (SPR). By comparison of a different TCR (B7) with similar affinity, and its binding to mutant peptide and mutant MHC complexes, it was shown that the two TCRs respond similarly to alterations in the MHC molecule but differ dramatically in their responses to ionic strength and temperature.
